Solanum viarum_H13

For RNA extractions we collected three mature leaves from all selected plants and collected root samples from a subset of 15 TA producing species and two Solanum species. The material was snap-freezed in liquid nitrogen and stored at -80C until extraction. For RNA extractions we pooled organ from the tree individuals to obtain a single pool per species. RNA was extracted with 1:1 trizolchloroform and precipitated in isopropanol and 75% ethanol. Quality control was performed by (1) electrophoresis in a 2% agarose gel, (2) evaluation of 260/280 and 260/230 absorbance ratios in a Nanodrop device, and (3) RNA quantification in Qubit. Samples that passed quality controls were sent to NovoGene for RNAseq genotyping using paired-end 150 bp libraries and an Illumina NovaSeq 6000 S4 Sequencing System.
