RNA-seq of Exocentrus adspersus: larval midgut

Larvae were sterilized by 70% ethanol and rinsed in double distillated water. Larvae were opened ventrally to expose the digestive tract and midgut were dissected out. Dissected gut were extensively washed in sterile water to remove gut content and frozen in liquid nitrogen and stored at -80C until further use. Total RNA was extracted from the dissected midguts using Trizol and DNA contamination was removed by DNAse treatment. Total RNA samples were purified by using RNeasy MinElute Clean up Kit and quantified using the RNA 6000 Nano LabChip kit on an Agilent 2100 Bioanalyzer. 150bp paired-end sequencing was performed on an Illumina Hiseq 2500 platform.
