Transcriptome of Diceraeus furcatus nymphs

Total RNA was extracted from whole bodies of ten first instar nymphs of each species using TRIzol Reagent (Invitrogen, Carlsbad, CA, USA), following manufacturers instructions. Total RNA was quantitated on a Nanodrop 2000c spectrophotometer (Thermo Scientific, Wilmington, USA) and its quality assessed with 1% (w/v) agarose gel electrophoresis. Immediately after extraction, 2 g of total RNA of each sample was lyophilized and shipped to Novogene (Davis, USA) for sequencing on an Illumina NovaSeq platform. The construction of cDNA libraries was performed from RNA samples for Illumina paired end (PE) sequencing following the Illumina protocol. The sequencing strategy was PE150, which gives 150bp read from each end to make a 300bp sequenced fragment.
