Three developmental stages were targeted: 1) fourth instar aphids (eleven days old), 2) reproductive young adult (fifteen days old) and 3) reproductive adult at peak fertility (twenty days old). Bacteriocytes were separated from the rest of the body with forceps under binocular loupe, collected with a peristaltic liquid pump and transferred in sterile plastic tubes before being immediately frozen at -80C prior RNA extraction.
