Total RNA was extracted using TRIzol, yielding 493 ng of RNA. Poly(A)+ RNA selection, TruSeq library preparation, and sequencing on a high-output run of an Illumina NextSeq 500 was performed by the Institute for Molecular Bioscience Sequencing Facility at The University of Queensland. This process yielded 26,482,164 150 bp paired-end reads, which were assembled de novo using an automated pipeline for trimming, correcting and assembling RNA-Seq reads using fqtrim, prinseq, and Trinity

RNA-Seq of Acharia sp. FG late instar venom spines

Tissue was stored in RNAlater reagent (Ambion). Total RNA was extracted from the venom spines and surrounding tissue using TRIzol (Thermo Fisher) according to the manufacturers instructions, with volumes reduced for smaller starting tissue masses. Poly-(A)+ enrichment and TruSeq library construction was performed by The University of Queenslands Institute for Molecular Bioscience (IMB) Sequencing Facility using either a NextSeq or HighSeq sequencer (Illumina).
